When you have a campaign in automation (Send campaign stage) the automation will listen to any unsubscribe lists, suppression lists, and dedicated campaign filters.

If your member passing through the automation is on the globally opt-out suppression list or unsubscribe list associated with the campaign or does not meet the filter criteria then they will be suppressed from the launch and then continue on the program.

TIP: You can run a test contact through the automation, i.e. add a contact to the automation, add them to the global opt-out unsubscribe list, progress them to a send campaign stage and check their events tab to see if they were suppressed.
